RESEARCHERS OF THE FGBI "VGNKI" INVESTIGATED THE INFLUENCE OF LASER RADIATION ON THE GENETIC MATERIAL OF BULLS
The scientific staff of the Rosselkhoznadzor subordinate fgbi "all-Russian state Center for quality and standardization medicines for animals and feed" (fgbi "VGNKI") established a negative influence of laser irradiation and fluorescent dye to the sperm quality of bulls. The results of this research make it possible to predict side effects of the separation of semen on the floor, widely used in farming today.

The study was conducted in the Department of the fgbi "VGNKI" quality control and standardization of genetic material and drugs used in animal reproduction. The results of the study were presented at the 22nd Annual conference of the European society for domestic animal reproduction ESDAR 2018 in late September and recently published in the scientific journal Reproduction in Domestic Animals included in the international database Web of Science.

Separation of semen on the floor, or sexyavenue seed –biotechnology, which from the beginning of the XXI century is used in animal breeding to produce offspring of the desired sex. The sex of the unborn calf depends on which sperm fertilizes the ovum of a cow. Sperm carry X or Y chromosomes. When fertilization of an egg by a sperm – bearer of the X chromosome is born a chick, a carrier of Y-chromosome – bull.

When sorting sperm by sex ejaculate spermatozoa separated into X - and Y-chromosomes in special installations – citosearch. The sperm are subjected to the staining effect of the laser radiation and pressure, electromagnetic influence and centrifugation. The negative effects of all these processes on the quality of sperm is not well understood, so research in this area is necessary to improve the separation technology, making it safer and more efficient.

Scientific employees of the Testing centre of the fgbi "VGNKI" conducted an experiment which studied the effect of diffuse and focused laser radiation and fluorescent dye Hoechst33342/PI in semen samples of cattle. For the experiment was constructed laser system with radiation parameters are identical to those used when sorting the seed on the floor on the production. Semen quality was investigated using Computer assisted semen analysis (CASA).

The result was to establish that the use of laser irradiation and staining with fluorochrome (Hoechst33342/PI) have a negative effect on sperm. Are the worst affected by such important factors as straightforwardly progressive motility and the number of fast cells. For example, at short-term laser irradiation of sperm (from 0.02 to 0.07 seconds) total speed of sperm is reduced by 7 times compared with the control samples.

According to experts of the fgbi "VGNKI", the study explains why in some cases the incoming samples of sperm, does not correspond to GOST standards.
